I’ve been working on a 6 month temporary position with an organization I really like. Recently, one of my coworkers left for grad school, so I’m applying to get into her position. I’d really love to get this job, but now I’m wondering about the tone a cover letter should take. I’ll be writing it to someone I know in HR, so it feels a little odd to be really formal and all, saying Dear Ms. Jane Smith and everything, when we’ve gone out for drinks with each other. Phrasing thoughts?

Generally err on the side of formality. Even if it feels stilted, it's appropriate, and projects that you know consider yourself a candidate like any other, personal affiliation notwithstanding. It shows you're taking it seriously. Adhering to the generally accepted standard won't be a bad thing.
